For the cleric i did maybe 8 zombie flesh trades and every other trade was unlocked. Sure, with the different enchantments you are right, but i still think its easier as in 1.7.

A villager becomes a cleric with a brewing stand. To lock their trades (and their profession) you just have to trade with them 1 time! Once you’ve traded with them, they will forever be a Cleric!

JEI is telling me that a level two cleric villager will trade ender pearls, but I have no idea how to level up villagers. Do I just have to have them loaded for long enough and it will just happen?

No unfortunately you can't. The only way to get Blaze Rods/Powder is from killing Blazes, so if your fortress doesn't have a spawner, you need to find another one.
